Gallbladder cancer (GBC) is a very aggressive tumor with a five-year survival rate under 15%. Worldwide GBC is considered a rare disease, but in Chile incidence rates amount to 25 cases per 100,000 person-years in women and 9 cases per 100,000 person-years in men. GBC caused 1.316 deaths in Chile in 2015 (931 women and 385 men). Current knowledge about the genetic and environmental triggers of this tumor is sparse. Gallstones disease is considered the main risk factor for CVB. Two observations support the important role of a genetic component that predisposes to these pathologies: (1) the broad association between ancestry and gallstones/GBC prevalence, and (2) the familial clustering observed for both diseases. However, genetic variants associated to GBC susceptibility in Chile have not yet been identified. Despite the large health impact of this cancer in Chile, few studies have been carried out in its population. In this sense, it is necessary to identify gallstones and GBC susceptibility markers in the Chilean population. The final objective of this thesis was to identify genetic variants associated with gallstones and GBC with familial clustering, and with the risk of sporadic GBC, through a three-step analysis: (1) genetic linkage analysis using parametric and non-parametric linkage methods in families with multiple GBC cases; (2) search for candidate genes through an integrative functional annotation analysis of the harbored genes in the identified linkage regions; and (3) fine-mapping by genetic association based on cases and population controls and using 1,028 variants distributed throughout the candidate regions. A total of 2,193 individuals were analyzed: 48 individuals (six cases of GBC, 15 cholecystectomized, 27 unaffected) belonging to 7 families with multiple GBC cases, 183 sporadic GBC patients and 1,962 population controls. Two candidate linkage regions were identified using non-parametric linkage analysis: Chr9: 105016960-115983216 and Chr16: 105444-5995388, which harbored 128 coding genes: 95 on chromosome 9 and 33 on chromosome 16. By functional annotation, the list was reduced to 30 genes potentially relevant for the disease etiopathogenesis. Candidate gene association analysis revealed two variants associated with sporadic GBC risk: ABCA1 rs363717 (OR = 1.84 95% CI 1.26-2.66) and SEC14L5 rs1558562 (OR = 2.02 95% CI 1.4-2.95). These results suggest that both GBC and gallstones with familial clustering, as well as sporadic GBC susceptibility in Chile, is associated at least in part with ABCA1 and SEC14L5 genetic variants....

The companion animals through the years have managed to position themselves in the family nucleus, being considered one more member of the family. Therefore, the death of the animal can be as significant as the loss of a loved one for its caregiver, bringing with it different consequences at a psychological, physical and social level. This project is part of a qualitative approach, focused on study of the grieving process by the loss of a companion animal and the generation development of products that respond to postulated requirements. The collection of relevant information was carried out through surveys on the subject and an exhaustive review of material and information from scientific articles and theses on Design, Psychology, Veterinary Medicine and Business Administration related to the relationship between pets and humans, process of bereavement and funeral rituals industry. As a result of the analysis carried out on the Theoretical Framework, State of the Art and Contextualization, this project obtains a design proposal that considers the realization of tangible symbolic elements of accompaniment during the grieving process of the loss of a pet and the realization of the respective prototypes to identify the most relevant materiality and production process for each case....
