Drugs of low aqueous solubility (BCS class II and IV) present a challenge to formulate and to be used in therapy. A new form of drug delivery is the printing of drugs in films for buccal administration. The problem for this is that the printing inks are aqueous based. Thus, these drugs also present problems to be formulated as inks. A possible solution to this is to encapsulate drugs in nanosystems or to nanonize the drug, to increase the amount of drug that can be suspended in the ink. The selected drug in this investigation was nifedipine, which has low aqueous solubility and it is also photosensitive. The nanosystems used were nanoemulsions, using the method of solvent displacement, and lipid core micelles, by the method of hot-melt emulsification. The nifedipine nanocrystals were developed using the high shear homogenization technique. Nanoparticles were characterized by size, PDI and zeta potential. Micelles and nanoemulsion were also observed under the microscope (TEM). Studies of stability, drug release and encapsulation efficiency were conducted. Once the nanosuspensions were characterized, the optimal formulations were mixed with glycerol in a 70:30 v/v ratio to develop the printing inks. The inks were printed and then, characterized in PDI size, zeta potential and printing efficiency. Nanocrystals of nifedipine were not viable for printing by the developed method, although, they achieved sizes suitable for being printed and avoided clogging the cartridge, only after a filtering process. Lipid core micelles, with sizes around 20 nm confirmed by microscopy, exhibited high encapsulation efficiencies. In the release study, they have shown a prolonged release profile and in terms of the stability, the formulations, both at room temperature and refrigerated, did not maintain their physical characteristics for more than a week. The nanoemulsion formulations have larger sizes and negative zeta potential. This probably gave greater stability than micelles, resulting in 4 weeks of physicochemical stability. The encapsulation efficiency was varied, but the drug loading was homogeneous among the formulations. Similar to micelles, nanoemulsions presented a prolonged release profile. In terms of printing, micelles and nanoemulsions printed a similar amount of nifedipine. Although micelles had higher encapsulation efficiencies, printing efficiency was lower than nanoemulsion. It was possible to print nifedipine inks, where the dispersion of the drug in the inks was increased. With nanoemulsions it was possible to disperse 160 times nifedipine versus its solubility in water. The micelles could disperse nifedipine more than 10 times at the end of the process...
