Genetics and the environment interact to generate a specific phenotype, which can be presented as an observable physical trait or complex behavior. The genetic variability within a population and its interaction with the environment generates different phenotypes. Prenatal nutrition is an environmental condition that has effects on physical traits, such as the small size of children with intrauterine growth restriction, and complex behaviors, such as sleep, memory and learning. However, despite the great impact that prenatal nutrition has on health and behavior, little is known about how it interacts with the genotype of individuals to influence their phenotype. Sleep is a very conserved behavior and highly variable within the population. Sleep disturbances such as chronic insomnia, sleep fragmentation and daytime sleepiness have been associated with mental illnesses such as bipolar disorder, Alzheimer's and Parkinson's. Therefore, it is very important to elucidate the genetic factors and mechanisms that underlie the relationship between these alterations and mental illness, in order to find new treatments. To answer this question, we used a subset of the Drosophila Genetic Reference Panel (DGRP) lines undergoing prenatal nutritional restriction. The DGRP is a collection of lines derived from a natural population, homozygous with isogenic and fully sequenced genomes, which represent the natural genetic variation within a population. Therefore, the genetic variation of DGRP can be associated with phenotypic variation using Genome-Wide Association Studies (GWAS). First, it was shown that genetic variation and its interaction with prenatal nutrition significantly impacts the variation in sleep behavior. Second, we found single nucleotide polymorphisms (Single Nucleotide Polymorphisms, SNPs) associated with the effect of prenatal nutrition on different sleep parameters, the associated genes were mapped, and with them a gene interaction network based on protein-protein interactions was generated. Within the network, groups of genes related mainly to the development of the organism were identified. We identified 110 (15%) candidate genes involved in nervous system development. Finally, we validated their role in sleep behavior by decreasing the function of 10 candidate genes belonging to this group. We found that all genes contributed to the genotype environment interaction since their loss of function has differential effects on each diet in at least one sleep variable. These results demonstrate the importance of nutritional genetic interaction during development and how different genes can affect adult sleep influenced by prenatal nutritional conditions...
