Stress precipitates a spectrum of neuropsychiatric disorders, including mood disorders, that is, complex and multifactorial entities characterized by a range of symptoms present in different combinations, suggesting the existence of stress-triggered disease subtypes. Using animal models, in our laboratory it was previously described that repetitive stress through two protocols: movement restriction, or immobilization, induced depressive-type behaviors in rats that were differentially reversed by antidepressant drugs that raise serotonin or norepinephrine levels, respectively. This suggests that the neurobiological mechanisms activated by both stressors are different. To characterize both stress protocols, we proposed to identify protein biomarkers in cerebrospinal fluid, finding that Aldolase C, an enzyme expressed in astrocytes, is present at differential levels in nanovesicular structures. Extracellular nanovesicles (ENVs) are secreted by all cell types, and they contain from proteins to miRNAs, and growing evidence points to their role in extracellular communication. The possibility that ENVs of brain origin could contain possible stress biomarkers would facilitate the diagnosis of complex psychiatric disorders. Therefore, we postulate the following hypothesis: ENVs of brain origin, obtained from serum, contain a differential protein profile in two models of chronic stress. To test our hypothesis, serum ENVs from rats exposed to stress were characterized, and their proteomes were identified by mass spectrometry. When validating the results, it was observed that Adolase C and the glial fibrillar acid astrocytic protein (GFAP) show differential levels in ENVs when comparing restriction with immobilization. To check whether Aldolase C in serum ENVs could be derived from forebrain cells, we transferred Aldolase C-GFP to astrocytes by in utero electroporation. Recombinant protein was recovered in serum ENVs, providing direct evidence that brain-derived ENVs are present in serum, while their molecular load could be a valuable source of biomarkers for central nervous system diseases...
