Shiga toxin-producing Escherichia coli (STEC) includes a group of foodborne pathogens that cause diarrhea and severe diseases, such as Hemorrhagic Colitis (HC) and Hemolytic Uremic Syndrome (HUS), primarily in children under 5 years of age. Shiga toxins are the major STEC virulence factors, but their ability to colonize the human intestine is a prerequisite to disease. The Locus of Enterocyte Effacement (LEE), a Pathogenicity Island (PAI), is a major determinant of intestinal epithelium attachment of a group of STEC strains (LEE-positive). The LEE-positive STEC strains (serogroups O26, O45, O103, O111, O121, O145 and O157) have been traditionally associated with outbreaks and HUS cases worldwide. On the contrary, in the case of STEC strains lacking LEE (LEE-negative), their virulence repertoire has not been fully characterized. It is noteworthy that the incidence of gastroenteritis caused by LEE-negative STEC strains (serogroups O91, O113, O128, O146 and O174) has increased in several countries, highlighting the relevance of their study. In this Thesis a novel PAI denominated Locus of Adhesion and Autoaggregation (LAA), which is exclusively present in LEE-negative STEC strains, is reported and characterized. Several virulence genes that participate in adhesion and colonization phenotypes are localized in LAA. In agreement with the above, the obtained results provide experimental evidence supporting the participation of LAA in the intestinal colonization of a mouse model of STEC infection. Additionally, more than 350 genomes of LEE-negative STEC strains were analyzed by comparative genomics and it was determined that LAA is the most frequent PAI in this subgroup of STEC. Through this analysis, other uncharacterized genomic islands were also identified, including two PAIs and one Integrative and Conjugative Element (ICE). Even more importantly, our phylogenetic analyzes indicate that in this subgroup of STEC there are geographically disseminated clades in which strains containing more than one PAI, including LAA, are clustered, which could contribute to the emergence of these pathogens. This study is a step forward in our knowledge of the evolution of genomes and pathogenic mechanisms of LEE-negative STEC, with important implications for their epidemiological surveillance....
