Estrogens are hormones of great importance that are associated with the female reproductive system and with the maintenance of secondary sex characteristics. These can be classified into two groups: (a) natural hormones, such as estrone (E1), estradiol (E2) and estriol (E3), together with its metabolites, and (b) synthetic hormones as 17α-ethinylestradiol (EE2), which is used in the formulation of contraceptive pills. Most natural estrogens are secreted by the kidney in their conjugated form as sulfates or glucuronides, but they can switch to the free estrogen. The quantification of this type of analytes in biological samples such as urine, can be a great challenge due to the low concentrations that they can be found, needing rigorous sample preparation steps, prior to analytical determination. In this work, an analytical strategy for determining estrogenic hormones: E1, E2, E3, its metabolites, and synthetic hormone EE2, from urine samples was developed based on the rotating disk sorptive extraction (RDSE) technique, and the subsequent detection and quantification by gas chromatography coupled to mass spectrometry (GC-MS). RDSE technique was selected due to its economic, versatile and eco-efficient (green chemistry) features, while GC-MS provides good sensitivity and low detection limits for to research in complex samples. The analytes are retained and pre-concentrated in the styrene-divinylbenzene (e-DVB) sorbent phase, which is immobilized on a rotating disk. The optimum conditions for extraction of all analytes were a rotation velocity of 3000 rpm, for 60 min and a sample volume of 25 mL (2 mL of urine and 23 mL of ultrapure water) at pH 7.0. Previous to the desorption step, a clean-up was performed after extraction with 5 mL of a methanol - water (1: 4) solution for 5 minutes. The desorption of the compounds was performed in one step of 15 minutes with 5 mL of methanol, then the extract was evaporated to dryness with nitrogen. An aliquiot of 50 μL of derivatizing agent N-methyl-N-(trimethylsilyl)-trifluoroacetamide (MSTFA) and 50 μL of pyridine were added to the dry eluate. Subsequently, it was subjected to a temperature at 90°C for 30 minutes. Once derivatization was complete, 20 μL of PCB 77 was added as internal standard and finally a volume of 2 μL was injected into the GC-MS. Furthermore, in the process, a standard surrogate ((20,21) -13C2-EE2) was incorporated. To know the total amount of estrogen present in the samples (conjugated + free) an enzymatic hydrolysis was performed, by treating a volume of 2 mL of urine with 2 mL of a buffer hydrolysis, containing the enzyme β-glucuronidase (responsible for deconjugation). This mixture of 4 mL was incubated for 20 hours at 37°C. After this time, the mixture was diluted to 25 mL with water and the extraction was carried out under the optimal conditions mentioned above. The detection and quantification limits of the method were between 0.057 to 0.71 μg·L-1 and 0.17 to 2.15 μg·L-1, respectively. Relative recoveries of blank urine samples that were spiked with 0.4 μg·L-1 of hormones were within a range of 67-98%. The described method was applied in 9 real samples from both women and men, in a wide age range (26-59 years). The concentration of the 9 hormone detected were obtained in a range of 0.5 to 366 μg·L-1and 1.1 to 709 μg·L-1, without and with hydrolysis, respectively...
