Non-alcoholic fatty liver disease (NAFLD) is multifactorial disease with complex interactions between diet, lifestyle and genetics. It has been found that is the most common cause of liver disease, making this a public health problem. It has been shown that insulin resistance (IR) and oxidative stress (OS) are the major determinants of the onset of NAFLD and progression to steatohepatitis. NAFLD is considered the hepatic manifestation of the metabolic syndrome and is directly related to the increased prevalence of obesity and type 2 diabetes mellitus (DM2). Currently, the only treatment is diet and changes in lifestyle. NAFLD patients show an increase in the parameters related to OS, IR and also liver reduction of long chain polyunsaturated fatty acids (LCPUFA) n-3, with increase in the ratio n-6/n-3. On the other hand, hydroxytyrosol (HT) is a polyphenol found mainly in olive products, which has potent antioxidant activity, protecting cells from OS and improving cardiac function. Given this evidence, we ask whether the use of LCPUFA n-3, specifically docosahexaenoic acid (DHA) plus HT, may have a protective effect against NAFLD and metabolic alterations that accompany it. Objective: To determine the effect of dietary supplementation with DHA and HT in reducing hepatic steatosis, inflammation and OS induced by high-fat diet in mice. Methodology: Male C57BL/6J mice were randomly assigned to each of the 8 experimental groups, where they were fed a control diet (CD, 10% of calories as fat) or a high-fat diet (HFD, 60% calories as fat) and supplemented with DHA (50mg/kg/day), HT (5mg/kg/day) or both for a period of 12 weeks. Results: HFD feeding for 12 weeks induced hepatic steatosis accompanied by OS and inflammation. In this regard, oral administration of DHA and HT achieved a greater protective effect than isolated supplementation of HT or DHA, compared to HFD through i) reduction of hepatic steatosis ii) less inflammatory response iii) increased antioxidant defenses and iv) increase the LCPUFA n-3 hepatic fatty acid profile. Conclusion: The HT + DHA mixed supplementation had a hepatoprotective role against injury caused by HFD, which could be considered as a possible clinical-nutritional intervention for handling NAFLD and its evolution steatohepatitis in mouse....

Evaluación farmacocinética de florfenicol en perros administrado por vía subcutánea

Actualmente los antimicrobianos disponibles para el tratamiento de cuadros respiratorios
en perros son limitados y requieren terapias prolongadas. En el presente estudio se estudiaron
distintos parámetros farmacocinéticos de florfenicol y florfenicol amina en perros, luego de la
administración de 10 mg/Kg del fármaco por vía subcutánea. Los resultados obtenidos para
florfenicol fueron los siguientes: concentración máxima (Cmax) de 1,58 ± 0,38μg/mL, tiempo en que se alcanzó la concentración máxima (Tmax) 3 ± 1,15 h, clearance (Cl) 1,01 ± 0,13 L/Kg/h, vida media (t½) 13,12 ± 3,10 h y volumen de distribución (Vd) 19,02 ± 3,87L/Kg entre otros valores. En cuanto a los parámetros registrados para florfenicol amina son los siguientes:
concentración máxima (Cmax) de 1,57 ± 0,36μg/mL, tiempo en que se alcanzó la concentración máxima (Tmax) 3 ± 1,15 h, clearance (Cl) 0,50 ± 0,21L/Kg/h, vida media (t½) 5,35 ± 1,82h y volumen de distribución (Vd) 4,22 ± 2,97L/Kg entre otros. Debido a que las concentraciones de florfenicol obtenidas en este estudio son más altas que las Concentraciones Mínimas Inhibitorias de las bacterias que afectan al sistema respiratorio de los animales, florfenicol podría ser una herramienta terapéutica para el tratamiento de patologías en perros....
