Introduction: Non-alcoholic fatty liver disease is the most important cause of chronic liver disease in the world. It is characterized by the abnormal accumulation of triacylglycerides in the hepatocytes (steatosis) that can evolve to inflammation, fibrosis and cirrhosis. The consumption of saturated fatty acids and excess carbohydrates, characteristic of the western diet, stimulate lipogenesis and hepatic steatosis, due to greater availability and oxidation of fatty acids, insulin resistance and decrease of long chain polyunsaturated fatty acids, n- 3 (LCPUFAs n-3), favoring a hepatotoxic and proinflammatory state. N-3 LCPUFAs, mainly eicosapentaenoic acid (C20: 5, EPA) and docosahexaenoic acid (C22: 6, DHA), control hepatic lipid metabolism by stimulating lipolysis and inhibiting lipogenesis, thus decreasing the inflammatory hepatic response. In this same line, Hydroxytyrosol (HT), has a marked cytoprotective effect and can prevent lipoperoxidation and oxidative DNA damage. Given these antecedents, the joint supplementation of DHA + HT suggests a synergy in the hepatoprotective effect. Objective: To evaluate the effect of dietary supplementation with DHA+HT on hepatic steatosis induced by a high-fat diet (HFD) by inactivating NF-κB and SREBP-1c. Methodology: Experimental, randomized, controlled, prospective study in animal model (male C57BL / 6J mice), randomly assignedto one of the eight experimental groups exposed to a control diet or high-fat diet (10% and 60% of calories from fat, respectively) with or without supplementation of DHA + HT. The presence of hepatic steatosis was evaluated by histology determined by the presence of fatty infiltration, the DNA binding activity of the transcription factors of SREBP-1c and NF-κB by ELISA and the hepatic expression of SREBP-1c mRNA, NF- κB, FAS, ACA, TNF-αand IL-1β by q-PCR. Results: Feeding with HFD induced hepatic steatosis, a proinflammatory and prolypogenic state. In opposition, the joint supplementation of DHA + HT in HFD produced i) less fatty infiltration in the liver; ii) decrease in the expression of proinflammatory markers TNF-α and IL-1β iii) decrease in the expression of prolypogenic enzymes FAS and ACA iv) Decrease in the activity of SREBP-1c and NF-κB Conclusions: The co-administration of DHA + HT can prevent the progression of NAFLD caused by HFD in mice by decreasing the activity of transcription factors SREBP1c and NF-κB and the proteins regulated and; FAS, ACA for SREBP1c and TNF-α and IL-1β for NF-κB....

Background: Nonalcoholic fatty liver disease (NAFLD) is the most common cause of liver disease. There is currently no drug therapy validated for NAFLD. Even though lifestyle interventions are the first-line therapy, low rates of adherence to this treatment have been reported. The n-3 long-chain polyunsaturated fatty acids (n-3 LCPUFA) have hepatoprotective effects. However, they are highly susceptible to lipid peroxidation and high doses have been associated with oxidative stress (OS) and disease progression. The addition of an antioxidant as a strategy to promote the beneficial effects of n-3 LCPUFA on NAFLD seems to be a reasonable approach. Objective: To evaluate the effect of combined supplementation of docosahexaenoic acid (DHA) with extra virgin olive oil (EVOO) in hepatic steatosis, OS, inflammation, metabolic disorders and insulin resistance induced by high fat diet (HFD) in mouse. Methods: C57BL/6J mice (n=11 per group) were fed for 12 weeks with a control diet (CD) (10% fat) or HFD (60% fat) and orally supplemented with saline (50mg/kg/day), DHA (50mg/kg/day), EVOO (50mg/kg/day) or EVOO + DHA (DHA 50mg/kg/day + EVOO 50mg/kg/day), thus eight experimental groups were established. After the intervention, the fatty acid profile in liver was determined by gas-liquid chromatography and parameters of hepatic steatosis, OS, inflammation and metabolic state in serum and liver tissue samples were measured by histological analysis, colorimetric and fluorometric kits, enzyme immunoassay, recycling enzymatic methodology and spectrophotometric methods. Results: HFD feeding for 12 weeks induced hepatic steatosis, OS and lost in the antioxidant capacity, systemic inflammation and metabolic disorders. Regarding supplementation with DHA plus EVOO resulted in greater protection than supplementation with only DHA or EVOO against HFD on i) reduction of hepatic steatosis; ii) reduced hepatic lipid-peroxidation; iii) low pro-inflammatory response and iv) lower insulin resistance, and it lead to the normalization of hepatic fat content, hepatic TAG and serum IL-6 in relation to the control group. Conclusion: Supplementation with DHA plus EVOO provided protection against injury caused by HFD; this way, it could be considered as a potential therapeutic line for handling NAFLD and prevention in the progression of nonalcoholic steatosis in mice....
