Background: Alzheimer’s disease (AD) is the most important form of dementia in the world population. The expression of amyloid precursor peptide (APP) in platelets has been postulated as a reliable biochemical marker for AD. Preliminary research done in our laboratory has shown that tau protein is also present in platelets and there are several tau fractions that can be resolved in electrophoresis. It has been suggested that tau fractions are altered in AD patients in an age independent manner. The present thesis aims to evaluate platelets tau fractions as a suitable biomarker for AD. -. Methods: Peripheral blood samples were obtained from 47 AD patients and 19 control subjects. Platelets were isolated and the tau patterns evaluated by immunofluorescence. Platelets proteins were analyzed by electrophoresis, immunoprecipitation and immunoblot techniques with tau and APP specific antibodies. The relationships between cognitive performance and platelets tau and APP expression in blood samples of subjects incorporated into the study were evaluated. -. Results: Platelets tau electrophoretic pattern showed the presence of different tau fractions. A higher ratio of high molecular weight forms and oligomers of tau was found in AD patients, with a direct correlation between platelets tau pattern and cognitive impairment. On the other hand, the analysis of platelets APP failed to demonstrate differences between controls and AD patients. -. Conclusions: We have confirmed the presence of tau protein in platelets of AD and control subjects. The electrophoretic pattern of platelets tau is compatible with the presence of high molecular weight aggregated forms of tau, which are increased in relation to other forms of tau in AD patients. There is a direct relationship between degree of the alterations in tau forms ratio and the degree of cognitive impairment of the subjects. No relationship was found between platelets APP electrophoretic pattern and cognitive status, or between APP pattern and tau forms ratio....
