La Enfermedad de Alzheimer (EA) es la principal causa de demencia en el adulto mayor, provocando deterioro cognitivo progresivo. Hoy en día no existe cura para esta enfermedad, además los tratamientos disponibles solo permiten retrasar el avance de la EA por un periodo de tiempo. Debido a esto se hace necesario crear un sistema para la detección temprana de la EA.
En esta memoria de Titulo se busca crear un sistema de nanopartículas que permitan mejorar la detección de agregados tóxicos del péptido β-amiloide (Aβ), mediante la técnica de espectroscopia de fluorescencia, ya que estos agregados están relacionados con el desarrollo de la enfermedad de Alzheimer (EA).
Para lograr esto se sintetizaron Nanovarillas de oro (NVO) y Nanoprismas de oro (NPrO), los cuales fueron estabilizados con polietilenglicol y posteriormente modificados con el péptido D1, este péptido reconoce selectivamente agregados del péptido Aβ. Estas nanopartículas fueron caracterizadas mediante espectrofotometría UV-Visible, dispersión dinámica de la luz (DLS), potencial Zeta (pZ) y microscopia electrónica de trasmisión y barrido (STEM). Se determinó que estas nanopartículas son capaces de aumentar la fluorescencia basal de la sonda fluorescente CRANAD-2 al estar unida a los agregados tóxicos del péptido Aβ, alcanzando un aumento de hasta 3 veces al utilizar NVO y de 2 veces al utilizar NPrO

Alzheimer's disease (AD) is the main cause of dementia in the elderly, causing progressive cognitive deterioration. Nowadays there is no cure for this disease, in addition the available treatments only allow to delay the advance of the AD for a period of time. Due to this it is necessary to create a system for the early detection of AD.
In this title report, the aim is to create a system of nanoparticles that can improve the detection of toxic aggregates of the β-amyloid peptide (Aβ), by means of the fluorescence spectrometry technique, since these aggregates are related to the development of the Alzheimer's disease (AD).
To achieve this, gold nanorods (NRO) and gold nanoprisms (NPrO) were synthesized, which were stabilized with polyethylene glycol and subsequently modified with the D1 peptide, this peptide selectively recognizes aggregates of the Aβ peptide. These nanoparticles were characterized by UV-Visible spectrophotometry, dynamic light scattering (DLS), Zeta potential (pZ) and electron microscopy of transmission and exploration (STEM). It was determined that these nanoparticles are able to increase the basal fluorescence of the fluorescent probe CRANAD-2 by being bound to the toxic aggregates of the Aβ peptide, reaching an increase of up to 3 times when using NRO and 2 times when using NPrO
