Lung cancer is the first cause of cancer-related deaths worldwide. Non-small cell lung cancer represents 85% of all lung cancer diagnosed worldwide, including a plethora of subtypes of cancer, namely adenocarcinoma and squamous cell carcinoma, among many others. Lately, several studies have confirmed the presence of Epstein-Barr virus in NSCLC cases. EBV is a herpesvirus associated with the development of many types of cancer in the human body, such as nasopharyngeal and gastric carcinoma and lymphomas. This have been related to the EBV capacity of developing a persistent infection and the establishment of a latency state in the human cells. In the context of a viral latency, it has been proved that EBV can undergo an abortive-lytic cycle, where the EBV expresses lytic cycle proteins but does not achieve cellular lysis. Among these lytic proteins, arises BamHI-A rightward frame 1 (BARF1), an early lytic protein with many notable properties, such as cell cycle promotion, apoptosis inhibition, cellular immortalization, and immune evasion. This protein has been exclusively found in EBV-related carcinomas. The hypothesis of this work establishes BARF1 as a protein that can promote phenotypical changes and oncogenesis in epithelial lung cells. The main goal of this work is to characterize those phenotypical changes in vitro, using healthy and cancerous lung cell models. Thereby, we transfected A549 and BEAS-2B cell lines to make them express BARF1 consistently, to study changes in cellular proliferation, migration and invasion, along with epithelial-mesenchymal transition. Finally, we studied the presence of EBV in Chilean NSCLC cases and their expression of BARF1. In this work we found that BARF1 expression correlated with increased cell proliferation, migration and invasion in vitro.
