Currently, there are a large number of chemical compounds considered emerging pollutants that can enter the environment, and exert negative effects on it, as is the case of antibiotics that can generate an increase in resistance to antimicrobials. Cefepime is one of the most widely used antibiotics in hospital care, which corresponds to a 4th-generation, high-spectrum cephalosporin used for the treatment of various infections, including those caused by Pseudomonas aeruginosa. Cefepime, as it is excreted mainly in the urine, enters the wastewater treatment plants of home and hospital origin. It is possible to find this compound in biosolids, a by-product of sewage treatment, which can be used as an organic amendment in agriculture, which could generate an increase in antimicrobial resistance in the surrounding biota. The objective of this research was to develop and validate an analytical methodology for the quantification of cefepime in urine matrix and biosolids from the Metropolitan Region of Chile. For the extraction of the analyte from the biosolid, two methodologies were considered, agitation and ultrasound. Liquid chromatography with diode array detector (HPLC-DAD) and gas chromatography with electron microcapture detector (GC-μECD) were used for quantification. The confirmation of the analyte in the matrices was carried out with the time-of-flight analyzer technique coupled to a mass detector with direct sample analysis (DSA-TOF-MS), which allows the analysis of the samples without prior treatment. Validation parameters such as sensitivity, precision, recovery and detection and quantification limits were evaluated for all the techniques used. The optimized working conditions for the analysis of the analyte in both matrices by means of HPLC-DAD, considered a C18 column of reversed phase, mobile phase of 90% CH3COOH 0.1M / 10% Acetonitrile. For the analysis by GC-μECD, an HP-5 column was used, with a 2-stage chromatographic program, performing the derivatization of the analyte with MSTFA. The results of the characterization of the biosolids used in this study showed that the organic carbon content was 72.9% for La Farfana and 48.3% for Melipilla. In relation to the extraction methodology of cefepime from biosolid, ultrasound turns out to be more efficient in relation to the necessary extraction time, obtaining recovery percentages close to 100%, with a precision of less than 0.75%, expressed as the standard deviation. relative. For the quantification of cefepime, the HPLC-DAD technique proved to be adequate for both matrices, with a recovery of 98.3% for urine and 96.3% for biosolids, with a retention time of 7.5 min, limits of detection and quantification less than 0.13 and 0.31 μg / mL, respectively. On the other hand, it was possible to develop a methodology to quantify MSTFA derivatized cefepime in ethyl acetate by GC-μECD, obtaining a retention time of 9.87 minutes with a recovery of 99.53%, a precision of less than 5.23%. and detection and quantification limits of 0.46 and 1.56 μg / mL, respectively. Finally, the presence of cefepime was confirmed using DSA-TOF-MS in the samples previously used in both HPLC-DAD and GC-μECD. The m / z ratio of 86 was identified, which belongs to the main metabolite of cefepime, N-methylpyrrolidine.
