Aging is the main risk factor associated to the development of chronic pathologies, including neurodegenerative diseases that compromises brain function. Prefrontal cortex is a brain region that experiments a functional decline during aging, which goes hand in hand with cognitive impairment, because of dampens in neuronal and synaptic activity. Several strategies focused into tackle the negative effects of aging in brain function have been proposed in the last years, ranging from pharmacological treatments to lifestyle changes, however they have not offered successful results. By contrast, recent studies show that nutritional interventions as ketogenic diet (KD), capable of inducing a metabolic change the brain is able to preserve memory in aging mice when is administrated in a cyclic way. This suggests that metabolic challenges could promote benefits on molecular components involved in synaptic function and neurotransmission in the brain cortex that has not been fully elucidated in detail. To address this hypothesis, the present work explored the molecular changes that KD elicits on prefrontal cortex of aged mice, by analyzing the presynaptic and postsynaptic cortical proteome when they were fed with cyclic KD at short-, medium- and long-term. Altogether, our results shows that cyclic KD has differential effects in the presynaptic and postsynaptic cortical proteome depending on the administration timeframe. Short-term treatment produces discrete changes in the postsynaptic component, whereas at medium-term and long-term administration, cyclic KD essentially modulates the presynaptic component, through key biological processes involved in synaptic function such as cAMP signaling pathway, actin cytoskeleton dynamics and synaptic vesicle trafficking. Against this background, we found that old mice preserved working memory which was correlated with higher dendritic complexity of pyramidal neurons in layers 2/3 of prefrontal cortex after twelve weeks of cyclic KD treatment. Finally, in vivo studies demonstrated that behavioral and structural changes were associated with increased kinase activity of protein kinase A (PKA) and high protein abundance of brain-derived neurotrophic factor (BDNF), components that play an essential role in development and synaptic plasticity maintenance. Overall, the findings obtained by this work contributes to elucidate the molecular mechanisms that cyclic KD regulates at some extent in vivo in the brain cortex, as result of metabolic switch induced by this intervention, which can preserve the cortical synaptic network during aging....
