Aging is the process in which optimal fitness declines over time. Neurons are cells with a primarily oxidative metabolism and together with astrocytes, counteract the reactive oxygen species (ROS) by-products. Astrocytes are a cell type of neuronal support, with antioxidant pathways that contribute to maintaining neuronal REDOX tone. There is evidence of changes in the expression of antioxidant proteins in aged astrocytes, but these studies fail to find a study model that accurately represents aging, given the difficulty of culturing astrocytes from adult animals. In this seminar, a cortical astrocyte primary culture protocol from young and old adult mice was successfully established in our laboratory. Furthermore, to understand astrocyte’s REDOX metabolism in aging, we integrated 4 aged mouse transcriptomes, focusing on astrocyte’s aging expression changes related to the metabolism of reactive oxygen species (ROS). Through gene ontology enrichment. (GO enrichment), analysis of KEGG pathways and STRING networks, we found changes in glucose metabolism, inflammatory and cell proliferation pathways. Also, crossing these data with the database of cysteine oxidation of the mouse proteome during aging (Oximouse), revealed that Ndufs1, is the protein with most sites number with oxidation percentage changes during aging, and that Prdx1, is the protein with the greatest change in oxidation percentage in aging. This show that in aging, there is specific changes in the expression and modification of ROS metabolism related proteins, that could impair REDOX homeostasis and astrocyte physiology....
