Preeclampsia (PE) is a specific multisystemic alteration of pregnancy, associated with hypertension, proteinuria and edema. Its etiology is still unknown, although it is related to a deficient placentation that leads to the development of exaggerated oxidative stress (OE) in the organ, accompanied by a greater apoptosis. To date and in relation to the placenta, it is important to emphasize that PE studies have been exclusively focused on the effect of OE and the apoptosis that the trophoblast undergoes, nevertheless a new cellular type in the stroma of the chorionic villi and have been termed Telocytes (TC). On the basis of the latter, this research focuses on the study of these new cells in cases of PE. The TCs present a strategic location between the myofibroblasts (MF) and the muscular layer of the great blood vessels of the chorionic villi, postulating that they operate as a functional triade in the regulation of the fetal vascular tone and the contraction-relaxation of the villi in the blood chamber, therefore they regulate the mother / fetus metabolic exchange. Since the placenta is a non-innervated organ, it is postulated that TC would act as a pacemaker or signal transducer for both the smooth muscle cells of fetal blood vessels and MF, both present in the villous stroma. Under the hypothesis that placentas from women with PE present higher OE and apoptosis evidenced by immunohistochemical markers, the objective of the study was to determine immunohistochemically in TC and MF the presence of nitrotyrosine residues as an OE marker and activated caspase 3 as a marker of the degree of apoptosis in these cells. Subsequently we analyze the relationship between these immunohistochemical cells parameters with gestational age, newborn weight (NB) and maternal age. Placenta samples (fetal side) were collected from preeclamptic (n=12) and normotensive patients (n=11). Standard immunoperoxidase techniques were individually applied in the tissue sections. Immunostaining was performed using streptavidin biotin kit (DAKO) according to manufacturer’s directions. The anti-nitrotyrosine A (Rabbit polyclonal Ab AB5411 Chemicon) was used in a 1: 100 dilution and the activated anti-caspase 3 Ab (monoclonal Ac CPP32 Novocastra), dilution 1:50, using diaminobenzidine as the chromogen. The immunohistochemical intensity of the reaction was evaluated by light microscopy (Zeiss Axioplan 2). The intensity of the immunohistochemical staining was measured using semi quantitative scores. Two randomly selected fields from each patient were evaluated by two different examiners who were blinded to the clinical information. The semi quantitative scoring of immunostaining intensity were assessed by the Pearson chi 2 test for differences between groups. Differences were considered statistically significant at p < 0.05. Chi-square statistic with p <0.05. As results it was observed that the TC and MF of the PE cases although they showed a greater expression of nitrotyrosine residues and activated caspase 3 than the control group, the differences were not statistically significant. Except for caspase 3 activated for MF, the only parameter that presented significant differences between groups. When the expression of nitrotyrosine and caspase 3 activated residues was related to the variables gestational age and NB, the results did not show significant differences between the groups, but independent of the clinical characteristic, all placentas were positive to the immunohistochemical antibodies used.
